Citotoxicidade de cimentos de ionômero de vidro restauradores sobre células de linhagem odontoblástica
Abstract
Os efeitos citotóxicos dos cimentos de ionômero de vidro podem ser atribuídos aos componentes liberados por estes materiais. Assim, o objetivo desta pesquisa foi avaliar a citotoxicidade dos extratos de um cimento de ionômero de vidro convencional (Ketac Fil Plus – 3M/ESPE) e de um cimento de ionômero de vidro modificado por resina (Fuji II LC – GC) aplicados sobre células da linhagem odontoblástica MDPC-23. Corpos de prova com dimensões padronizadas foram preparados, imersos em 1,1mL de meio de cultura (DMEM), e incubados por 24 horas em estufa a 37ºC com 5% de CO2. Os extratos obtidos foram filtrados, tiveram o pH ajustado para 7,4 e, foram incubados por 24 horas em contato direto com as células MDPC-23. DMEM puro foi utilizado como controle negativo. O metabolismo celular foi determinado pelo teste do MTT e os valores numéricos obtidos foram submetidos à análise estatística. A morfologia das células em contato com os extratos dos materiais experimentais ou controle (DMEM) foi avaliada em microscopia eletrônica de varredura (MEV). Os resultados obtidos pelo teste do MTT não mostraram diferença estatística significante entre os dois materiais e o grupo controle. Na análise em MEV, tanto para os cimentos de ionômero de vidro como para o grupo controle, foi observado um grande número de células com morfologia ligeiramente alongada e com múltiplos prolongamentos citoplasmáticos. Assim, foi possível concluir que os materiais ionoméricos avaliados apresentam baixo efeito citotóxico para as células de linhagem odontoblástica MDPC-23.

SUMMARY
The cytotoxic effects of glass-ionomer cements (GICs) may be attributed to the compounds released from these biomaterials. Consequently, the aim of this in vitro study was to evaluate the cytotoxicity of the extracts obtained from a conventional glass-ionomer cement (Ketac Fil Plus – 3M/ESPE) and a resin-modified glass-ionomer cement (Fuji II LC – GC) applied to an immortalized odontoblast cell line (MDPC-23). Standardized round-shaped samples were prepared, immersed in 1.1 mL of culture medium (DMEM), and incubated for 24h at 37ºC with 5% CO2 and 95% air. The extracts from the experimental materials were filtered and the final pH values were adjusted to 7.4. These extracts were applied on the MDPC-23 cells and incubated for additional 24 hours. Fresh DMEM was used as the control group. The cell metabolic activity was determined by the MTT assay and the data were submitted to the statistical analysis. Cell morphology was evaluated by scanning electron microscopy (SEM). The results of the MTT assay determined no statistical difference among the experimental and control groups. In the cell morphology assessment, a number of cells with spindle shape and several cellular processes on their cytoplasmatic membrane were showed for all groups. It was concluded that both glass-ionomer cements evaluated present low cytotoxic effects to the MDPC-23 cells.
